Research has unveiled a potential breakthrough in reversing a specific type of hearing loss. A recent study indicates that mutations in a particular gene, occurring at birth, may be effectively countered by a widely used supplement in conjunction with the erectile dysfunction medication Viagra, known scientifically as sildenafil.
The study focused on a gene called CPD, which produces the enzyme carboxypeptidase D. This gene was found to harbor mutations in three unrelated families from Turkey, each experiencing cases of congenital deafness.
The identified families suffered from a form of inherited sensorineural hearing loss. This condition results from damage to the delicate hair cells in the inner ear responsible for transforming sound vibrations into electrical signals that the brain interprets. This detrimental effect has been documented by the National Institutes of Health.
To deepen their understanding, researchers compared genetic information from affected and unaffected relatives within these families, subsequently expanding the scope of their analysis to include other populations.
According to Dr. Mustafa Tekin, a leading investigator and professor at the University of Miami Miller School of Medicine, the team discovered CPD mutations present among individuals with hearing loss in a substantial genome sequencing cohort from England.
During laboratory experiments, scientists inactivated the CPD gene in human cells. They observed significant reductions in levels of three critical substances: arginine, an amino acid; cyclic guanosine monophosphate, known as cGMP, which functions as an essential signaling molecule; and nitric oxide, crucial for transmitting vital signals throughout the nervous system.
The initial phases of the study utilized human test subjects, but the team subsequently expanded its research to include experiments conducted on mice and fruit flies.
When researchers noted diminished nitric oxide and cGMP levels, they found that the inner ear hair cells in mice faced overstress, ultimately dying. Similarly, in fruit flies, silencing the equivalent CPD gene led to impairments in the organ essential for sound detection and balance.
However, when arginine was administered to human cells, it restored nitric oxide levels to normal, resulting in reduced cell death. Moreover, Viagra, known for its effects on the nitric oxide pathway, demonstrated the capability to reverse some deficits caused by CPD mutations, Dr. Tekin added.
This research suggests that a deficiency of nitric oxide in the cochlea, which is the part of the inner ear that converts sound vibrations into nerve signals, may significantly contribute to hearing loss. The CPD gene plays a vital role in regulating this process by maintaining optimal levels of arginine and nitric oxide in hair cells, both of which are essential for proper signal transmission and protection against noise-induced damage.
Dr. Tekin emphasized the importance of maintaining a delicate balance of nitric oxide for the health of various tissues.
Despite these promising findings, the researchers have clarified that they do not recommend Viagra as a treatment for hearing loss.

Experts outside of the research team also cautioned about potential adverse effects associated with Viagra. It is crucial to note that, at this point, it is neither advisable nor safe for the general population to use Viagra as a remedy for hearing issues, Dr. Parhizkar asserted.
Research has shown that the hearing loss resulting from Viagra usage typically proves to be temporary and reversible upon discontinuation of the medication.
The Food and Drug Administration has included a warning label on Viagra and other PDE5 inhibitors regarding the possible risk of sudden hearing loss following rare reports among individuals using this class of drugs.
Experts remain uncertain whether this side effect arises directly from PDE5 inhibitors since affected patients may have other underlying medical conditions or could be taking additional medications that might contribute to hearing loss.
Given that the study concentrated on a very small, genetically distinct group of patients, Dr. Parhizkar noted that its findings are unlikely to be broadly applicable to the general population.
With the research also assessing hearing loss reversal in animal models, further studies are essential to evaluate potential treatments in humans.
Dr. Tekin advised individuals experiencing any hearing loss symptoms to undergo genetic testing to identify the underlying causes. Although scientists have pinpointed over 200 genes associated with hearing loss, existing commercial genetic screening tests often target only the most prevalent mutations.
